In this introductory article to the special issue on Multi-level semiosis we attempt to stage the background for qualifying the notion of “multi-levelness” when considering communication processes and semiosis in all life forms, i.e. from the cellular to the organismic level. While structures are organized hierarchically, communication processes require a kind of processual organization that may be better described as being heterarchical. Theoretically, the challenge arises in the temporal domain, that is, in the developmental and evolutionary dimension of dynamic semiotic (...) processes. We discuss the importance of this fundamental difference in order to explain how levels, domains and orders of magnitude, on the one hand, and synchronic and diachronic processes, on the other, contribute to the overall organization of every living being. To account for such multi-level organization, semiotic freedom is assumed to be a scalar property that endows living systems at different levels and domains with the capacity to ponder selectively the overall structural coherence and functional compatibility of their heterarchical processing, which is increasingly less conditioned by the underlying molecular determinism. The majority of G protein-coupled receptors (GPCRs) self-assemble in the form dimeric/oligomeric complexes along the plasma membrane. Due to the molecular interactions they participate, GPCRs can potentially provide the framework for discriminating a wide variety of intercellular signals, as based on some kind of combinatorial receptor codes. GPCRs can in fact transduce signals from the external milieu by modifying the activity of such intracellular proteins as adenylyl cyclases, phospholipases and ion channels via interactions with specific G-proteins. However, in spite of (...) the number of cell functions they can actually control, both GPCRs and their associated signal transduction pathways are extremely well conserved, for only a few alleles with null or minor functional alterations have so far been found. This would seem to suggest that, beside a mechanism for DNA repairing, there must be another level of quality control that may help maintaining GPCRs rather stable throughout evolution. We propose here receptor oligomerization to be a basic molecular mechanism controlling GPCRs redundancy in many different cell types, and the plasma membrane as the first hierarchical cell structure at which selective categorical sensing may occur. Categorical sensing can be seen as the cellular capacity for identifying and ordering complex patterns of mixed signals out of a contextual matrix, i.e., the recognition of meaningful patterns out of ubiquitous signals. In this context, redundancy and degeneracy may appear as the required feature to integrate the cell system into functional units of progressively higher hierarchical levels. (shrink)

Any living organism interacts with and responds specifically to environmental molecules by expressing specific olfactory receptors. In this paper, this specificity will be first examined in causal terms with particular emphasis on the mechanisms controlling olfactory gene expression, cell-to-cell interactions and odor-decoding processes. However, this type of explanation does not entirely justify the role olfactory receptors have played during evolution, since they are also expressed ectopically in different organs and/or tissues. Homologous olfactory genes have in fact been found in such (...) diverse cells and/or organs as spermatozoa, testis and kidney where they are assumed to act as chemotactic sensors or renin modulators. To justify their functional diversity, homologous olfactory receptors are assumed to share the same basic role: that of conferring a self-identity to cells or tissues under varying environmental conditions. By adopting this standpoint, the functional attribution as olfactory or chemotactic sensors to these receptors should not be seen either as a cause conditioning receptor gene expression, or as a final effect resulting from genetically predetermined programs, but as a direct consequence of the environmental conditions olfactory receptor genes have explored during evolution. The association of odorant patterns with specific environmental or contextual situations makes their relationship semiotically triadic, due to the emergence of an interpretant capable of perceiving odorants as meaningful signs out of a noisy background. This perspective highlights the importance of odorant-receptor relationships as respect to the properties of the interacting partners. It is our contention that only when taken together can these different explanatory strategies provide a realistic account of how olfactory receptor genes have been structurally and functionally modified during evolution. Receptor oligomerization plays a key role in maintaining genome stability and restricting protein mutagenesis. When properly folded, protein monomers assemble as oligomeric receptors and interact with environmental ligands. In a gene-centered view, the ligand specificity expressed by these receptors is assumed to be causally predetermined by the cell genome. However, this mechanism does not fully explain how differentiated cells have come to express specific receptor repertoires and which combinatorial codes have been explored to activate their associated signaling pathways. It is (...) our contention that the plasma membrane acts as the locus where several contextual cues may be integrated. As such it allows the semiotic selection of those receptor configurations that provide cells with the minimum essential requirements for agency. The occurrence of protein misfolding makes it impossible for receptor monomers to assemble along the membrane and to sustain meaningful relationships with environmental ligands. How could a cell lineage deal with these loss-of-function mutations during evolution and restrain gene redundancy accordingly? In this paper, we will be arguing that the easiest way for bacteria clones to accomplish this goal is by getting rid of cells expressing mutated receptor proteins. The mechanism sustaining this cell selection is also occurring in many somatic tissues and its function is currently believed to counteract in vivo protein mutagenesis. Our discussion will be mainly focused on the significance and semiotic nature of the interplay between membrane receptors and the epigenetic control of gene expression, as mediated by the control of mismatched repairing and protein folding mechanisms. Germ cells are cross-roads of development and evolution. They define the origin of every new generation and, at the same time, represent the biological end-product of any mature organism. Germ cells are endowed with the following capacities: to store a self-descriptive program, to accumulate a protein-synthesizing machinery, and to incorporate enough nourishment to sustain embryonic development. To accomplish this goal, germ cells do not simply unfold a pre-determined program or realize a sole instructive role. On the contrary, due to the (...) complexity of their cytoarchitecture and the nature of the soma-to-germ interactions, they are heavily involved in processes of sign recognition and meaningful tissue exploration. At each stage of their inward migration, germ plasma membranes act as semiotic interfaces allowing cells to interact with the surrounding extracellular milieu and experience the compatibility of selected developmental sequences. The question of which signaling pathways are activated at each developmental stage does not result from a strictly predetermined program instructing germ cell stemness. Rather, each developmental sequence is an open-ended semiotic relationship explored and gradually defined during evolution by the context-dependency of specific cell-to-cell interactions. In this way, any structural and functional novelty that has emerged in the course of germ cell interactions may be interpreted as an exaptation fixed in the species genome in response to specific environmental constraints. The egg behaves as a prospective cell sustaining the developmental processes of the future embryo. In biosemiotic terms, this apparent teleonomic behaviour can be accounted for without referring to the exclusive causal role played by its genetic makeup. We envision two different processes that are uniquely found in the oocyte: (1) the first involves the mechanisms by which large amounts of mRNA accumulate in the ooplasm to establish the embryo axes prior to fertilization; (2) the second involves transfer of an (...) excess of maternally supplied ribosomes to the oocyte to provide the future embryo with newly synthesized proteins. In this paper, we argue that the information required to sustain embryonic development is not due to any physical properties of the zygotic DNA and the maternal mRNAs, but to their spatially and temporally ordered relationship in the zygote’s internal space. Franco Eugeni remembers Bruno Rizzi: in this brief introduction, I would like to remember an afternoon spent in “ Roma Tre ” with Bruno, since we were both Ordinary Professors at that University. We passed it doing a dense program of work for the next three years. At 6.00 pm, I left for “Roseto degli Abruzzi”. At six o'clock a.m. of the next morning, I still have the voice in my ears. A phone call from the Headmaster Ciro d'Aniello, (...) who told me " The Professor is dead" In that afternoon, Emilio Ambrisi and I went to Rome; we were stunned and desperate. Not many days later, a conference was held at the University of Teramo; it was the conference that we had prepared for Bruno, to celebrate his 60th birthday, but it was his memory. In the opening conference, Prof. Antonino Giambo, also fraternal friend of Bruno, burst into a tearful cry, which expressed in a moment, the senses of friendship and love that we all had for the missing friend! Starting from this work with Fabrizio Maturo, the study of some problems left open by the works of Eugeni and Rizzi will be investigated. Sunto Ricordo di Franco Eugeni su Bruno Rizzi: in questa brevissima introduzione vorrei ricordare un pomeriggio passato a Roma Tre con Bruno, visto che allora eravamo entrambi Professori Ordinari in quella Università. Lo passammo a fare un denso programma di lavoro per i successivi tre anni. Alle 18.00 ripartii per Roseto degli Abruzzi. Alle sei del mattino successivo, ho ancora la voce nelle orecchie, una telefonata del Preside Ciro d’Aniello, che mi diceva “ Il Professore è morto” Nel pomeriggio Emilio Ambrisi ed io ci recammo a Roma, eravamo attoniti e disperati. Non molti giorni dopo si tenne presso l’Università di Teramo un Convegno, era il Convegno che avevamo preparato per Bruno, per festeggiare i suoi 60 anni, fu invece il ricordo. Nella conferenza di apertura il prof. Antonino Giambò, anche lui, come noi, fraterno amico di Bruno, scoppiò in un pianto dirotto, che espresse in un attimo, i sensi dell’amicizia e dell’amore, che tutti noi avevamo per l’amico scomparso! A partire da questo lavoro con Fabrizio Maturo verrà approfondito lo studio di alcune problematiche rimaste aperte dai lavori di Eugeni e Rizzi. (shrink)

Este artículo establece relaciones entre el mensaje de algunos manuales de Historia del bachillerato franquista, utilizados como fuentes primarias (29 en total) y la posición política de sus autores, fueran castigados (depuración en el escalafón de catedráticos de 1935), o premiados con la convocatoria de oposiciones patrióticas de 1940 que favorecieron a los simpatizantes del golpe de Estado de 1936. Para descubrir esas trayectorias personales se recurre a fuentes secundarias. Si unos autores utilizan el manual de Historia para constatar su (...) fidelidad personal al régimen de Franco; otros, lo utilizan como salvoconducto, para marcar distancias con su pasado masón, republicano de izquierdas, catalanista. Si en autores como José Ramón Castro hay coherencia entre su producción historiográfica profesional y escolar (su fidelidad al régimen es inquebrantable), Jaime Vicens Vives o Antonio Domínguez Ortiz, como autores de manuales representan la cultura oficial (legitiman al régimen); como historiadores profesionales, la denominada cultura establecida, al distanciarse más de él y de sus valores. In this detailed study of the republican tradition in the development of the Enlightenment, the central problem of utopia and reform is crystallized in a discussion of the right to punish. Describing the political situation in Europe in the seventeenth and eighteenth centuries, the author shows how the old republics in Italy, Poland and Holland stagnated and were unable to survive in the age of absolutism. The Philosophes discussed the ideal of republicanism against this background. They were particularly influenced by (...) the political and religious radicalism of John Toland, which had survived the English Restoration and was then reaching Europe. Professor Venturi traces the debate on the penal laws and the attempt to relate utopian ideas of society to the practical problem of dealing with man in society, which culminated in the assertion by many Philosophes that an unjust social system necessitated harsh penal laws, thereby rejecting the possibility of reform. Le présent ouvrage comble une lacune. La recherche, en effet, n'avait jamais pris en vue de manière systématique et détaillée l'ensemble de la philosophie schellingienne dans son rapport à la théologie. (C'est d'autant plus surprenant si l'on se rappelle que la pensée de Schelling représente le dernier essai de grand style d'établir une médiation entre ces deux disciplines.) En pratiquant sur toute l'œuvre une coupe longitudinale - depuis les premiers écrits critiques sur le mal et le mythe, à travers la (...) spéculation théogonique des Ages du monde, jusqu'aux développements christologiques et ecclésiologiques de la dernière période - le livre retrace le devenir de la théologie de Schelling, s'intégrant dans sa pensée comme l'une de ses composantes. Cette méthode permet de mettre en évidence la façon dont le philosophe théologien s'est critiqué lui-même dans les diverses phases de sa longue carrière et a modifié ses positions initiales pour tenter d'échapper à la synthèse idéaliste de la foi et du savoir, sans parvenir toutefois à se dégager entièrement du schéma de l'identité. Grace a son interrogation incomparablement tenace et incisive sur le sens de l'etre, la pensee heideggerienne accede a une comprehension du sacre qui semble irreductible a tout autre, et qui, par son originalite et sa profondeur, compte parmi les plus significatives elaborees a notre epoque. Le present ouvrage etudie le rapport complexe entre cette notion du sacre et les eclaircissements heideggeriens sur le divin et la mediation poetique. Il s'articule en quatre parties, dont la premiere expose la genese de l'interpretation (...) heideggerienne du sacre. La deuxieme partie propose une etude thematique du nouveau lexique que Heidegger, au contact notamment de la poesie de Holderlin, introduit dans sa meditation du sacre. La troisieme partie entreprend une serie de discernements critiques, en se laissant instruire par les principaux travaux qui ont examine explicitement la comprehension heideggerienne du sacre, du divin, et du poete. Dans une perspective synthetique, la quatrieme partie, enfin, s'efforce de ressaisir l'approche de Heidegger par le biais de ce qui la deborde, en la faisant dialoguer avec d'autres pensees philosophiques et theologiques marquantes; ce qui conduit a considerer, entre autres, la difference du sacre et du Saint. Degage a partir des developpements diachroniques de la premiere partie, le triptyque du sacre, du divin, et du poete structure aussi bien les synchronisations thematiques de la deuxieme partie que les discernements critiques fournis par la troisieme, et il est assume, tout en etant "depasse", par les chapitres systematiques de la derniere partie. We present a preliminary high-level formal theory, grounded on knowledge representation techniques and foundational ontologies, for the uniform and integrated representation of the different kinds of (quali- tative and quantitative) knowledge involved in the designing process. We discuss the conceptual nature of engineering design by individuating and analyzing the involved notions. These notions are then formally charac- terized by extending the DOLCE foundational ontology. Our ultimate purpose is twofold: (i) to contribute to foundational issues of design; and (ii) to support (...) the development of advanced modelling systems for (qualitative and quantitative) representation of design knowledge. Product structures are represented in engineering models by depicting and linking components, features and assemblies. Their understanding requires knowledge of both design and manufacturing practices, and yet further contextual reasoning is needed to read them correctly. Since these representations are essen- tial to the engineering activities, the lack of a clear and explicit semantics of these models hampers the use of information systems for their assessment and exploita- tion. We study this problem by identifying different interpretations of structure rep- resentations, (...) and then discuss the formal properties that a suitable language needs for representing components, features and combinations of these. We show that the representation of components and features require a non-standard mereology. (shrink)

Background:A growing number of frameworks for the practice of clinical ethics are described in the literature. Among these, hermeneutical frameworks have helped highlight the interpretive and contextual nature of clinical ethics practice.Objectives:The aim of this article is to further advance this body of work by drawing on the ideas of Charles Taylor, a leading hermeneutical philosopher.Design/Findings:A Hermeneutical Rapprochement Framework is presented for clinical ethics practice, based on Taylor’s hermeneutical “retrieval” and “rapprochement.” This builds on existing hermeneutical approaches for the practice (...) of clinical ethics by articulating a framework with interpretive and reconciliatory scope that extends beyond the presenting “local” context. A Hermeneutical Rapprochement Framework considers broader socio-historical horizons and imaginaries grounded on Taylor’s expansive work in epistemological, ontological, political, and moral philosophy.Discussion:The framework is discussed in terms of how it can be operationalized for clinical practice as well as normative development. Implications for the educational preparation of clinical ethicists are also discussed. Although this work is directly relevant for clinical ethicists, it can also help inform the ethical practice of all clinicians. (shrink)
